HanchorBio Announces Acceptance of Four Scientific Abstracts for ESMO Congress 2026, Highlighting Differentiated Immunotherapy Strategy
HCB101’s engineered macrophage activation and emerging clinical evidence support its potential as a combination backbone, while HCB301 demonstrates the multi-specific engineering capabilities of HanchorBio’s FBDB™ platform
TAIPEI, SHANGHAI, and SAN FRANCISCO — October 27, 2026 — HanchorBio, Inc. (TWSE: 7827), a global clinical-stage biotechnology company developing next-generation immunotherapies for oncology and immune-mediated diseases, today announced that four scientific abstracts covering HCB101 and HCB301 were accepted for ESMO Congress 2026, taking place October 23–27 in Madrid, Spain.
The research highlights HanchorBio’s strategy to advance immunotherapy beyond single-pathway checkpoint blockade by integrating macrophage-mediated tumor clearance with established and emerging cancer treatments.
HCB101: Beyond CD47 Blockade — Engineered for Combination
HCB101 is an engineered SIRPα–IgG4 Fc fusion protein designed to block the CD47–SIRPα innate immune checkpoint while minimizing red blood cell binding.
Unlike CD47 inhibitors incorporating an effector-silenced IgG1 Fc, HCB101 retains macrophage-mediated phagocytic activity through its functional IgG4 Fc-containing architecture. Published preclinical studies demonstrated robust tumor-cell phagocytosis with minimal red blood cell phagocytosis.
This functional distinction provides a scientific rationale for combining HCB101 with therapies that do not provide strong Fc-mediated tumor-opsonizing activity, including PD-1 inhibitors, chemotherapy, anti-angiogenic agents, and HER2/EGFR-directed antibodies.
Emerging clinical evidence supports further evaluation of this strategy. HCB101 monotherapy demonstrated sustained CD47 receptor occupancy and a confirmed partial response in head and neck cancer, with treatment ongoing beyond 83 weeks. In second-line gastric/gastroesophageal junction (GC/GEJ) cancer, HCB101 combined with ramucirumab and paclitaxel achieved a preliminary objective response rate (ORR) of 50.0% (11/22) and disease control rate (DCR) of 100% (22/22). Additional activity was observed in first-line HER2-positive gastric cancer (ORR 77.8%, 7/9) and second-line colorectal cancer (ORR 35.3%, 6/17).
These early, nonrandomized clinical findings support continued evaluation of HCB101 across distinct treatment backbones, with controlled studies needed to establish its incremental therapeutic contribution. Second-line GC/GEJ cancer remains HanchorBio’s lead clinical development priority.
From Clinical Evidence to Partner-Enabled Innovation
Building on this clinical foundation, HanchorBio is exploring next-generation combinations involving PD-1/VEGF bispecific antibodies, antibody-drug conjugates (ADCs), therapeutic cancer vaccines, and small molecules targeting complementary tumor microenvironment pathways.
Its collaboration strategy includes InxMed’s FAK inhibitor IN10018 and a recently signed memorandum of understanding with Aston Sci. involving therapeutic cancer vaccines.
These initiatives reflect HanchorBio’s ambition to develop HCB101 as a versatile macrophage-directed backbone that may complement therapeutic innovation beyond its internal pipeline.
“The next generation of immunotherapy must do more than release inhibitory checkpoints. It must engage complementary immune mechanisms that can work together,” said Scott Liu, PhD, Founder, Chairman, and CEO of HanchorBio. “With HCB101, we have engineered a molecule that goes beyond CD47 blockade to support macrophage-mediated tumor clearance while minimizing red blood cell interactions. Our emerging clinical evidence supports development across multiple treatment combinations, from established standards of care to bispecific antibodies, ADCs, and cancer vaccines. Through our FBDB™ platform and expanding collaborations, we aim to translate this differentiated biology into broader therapeutic opportunities.”
FBDB™: Extending Innate Immunity Through Multi-Specific Engineering
HanchorBio’s proprietary Fc-Based Designer Biologics (FBDB™) platform combines protein engineering, Fc-function optimization, and multi-specific molecular design to develop therapies addressing complementary immune mechanisms.
HCB301 exemplifies this approach through a trispecific fusion protein that integrates SIRPα, PD-L1, and TGFβ pathway modulation into a single molecule, designed to address innate immune suppression, adaptive checkpoint resistance, and tumor microenvironment regulation.
A scientific poster featuring early HCB301 Phase 1 findings will be presented by a study investigator at ESMO Congress 2026. The study’s clinical safety findings and therapeutic window require further evaluation.
